    ShopSmart 4 Health - protocol of a skills-based randomised controlled trial promoting fruit and vegetable consumption among socioeconomically disadvantaged women

    BackgroundThere is a need for evidence on the most effective and cost-effective approaches for promoting healthy eating among groups that do not meet dietary recommendations for good health, such as those with low incomes or experiencing socioeconomic disadvantage. This paper describes the ShopSmart 4 Health study, a randomised controlled trial conducted by Deakin University, Coles Supermarkets and the Heart Foundation, to investigate the effectiveness and cost-effectiveness of a skill-building intervention for promoting increased purchasing and consumption of fruits and vegetables amongst women of low socioeconomic position (SEP).Methods/designShopSmart 4 Health employed a randomised controlled trial design. Women aged 18&ndash;60 years, holding a Coles store loyalty card, who shopped at Coles stores within socioeconomically disadvantaged neighbourhoods and met low-income eligibility criteria were invited to participate. Consenting women completed a baseline survey assessing food shopping and eating habits and food-related behaviours and attitudes. On receipt of their completed survey, women were randomised to either a skill-building intervention or a wait-list control condition. Intervention effects will be evaluated via self-completion surveys and using supermarket transaction sales data, collected at pre- and post-intervention and 6-month follow-up. An economic evaluation from a societal perspective using a cost-consequences approach will compare the costs and outcomes between intervention and control groups. Process evaluation will be undertaken to identify perceived value and effects of intervention components.DiscussionThis study will provide data to address the currently limited evidence base regarding the effectiveness and cost-effectiveness of skill-building intervention strategies aimed at increasing fruit and vegetable consumption among socioeconomically disadvantaged women, a target group at high risk of poor diets.<br /

    Spatially Resolved Far-Ultraviolet Spectroscopy of the Nuclear Region of NGC 1068

    We carry out high-resolution FUSE spectroscopy of the nuclear region of NGC 1068. The first set of spectra was obtained with a 30" square aperture that collects all emission from the narrow-line region. The data reveal a strong broad OVI component of FWHM ~3500 kms-1 and two narrow OVI 1031/1037 components of ~350 kms-1. The CIII 977 and NIII 991 emission lines in this spectrum can be fitted with a narrow component of FWHM ~1000 kms-1 and a broad one of ~2500 kms-1. Another set of seven spatially resolved spectra were made using a long slit of 1.25" X 20", at steps of ~1" along the axis of the emission-line cone. We find that (1) Major emission lines in the FUSE wavelength range consist of a broad and a narrow component; (2) There is a gradient in the velocity field for the narrow OVI component of ~200 kms-1 from ~2" southwest of the nucleus to ~4" northeast. A similar pattern is also observed with the broad OVI component, with a gradient of ~3000 kms-1. These are consistent with the HST/STIS findings and suggest a biconical structure in which the velocity field is mainly radial outflow; (3) A major portion of the CIII and NIII line flux is produced in the compact core. They are therefore not effective temperature diagnostics for the conical region; and (4) The best-fitted UV continuum suggests virtually no reddening, and the HeII 1085/1640 ratio suggests a consistently low extinction factor across the cone.Comment: To appear in the Astrophysical Journal. 37 pages with 12 figure

    Reliability of the biceps brachii M-wave

    BACKGROUND: The peak-to-peak (P-P) amplitude of the maximum M-wave and the area of the negative phase of the curve are important measures that serve as methodological controls in H-reflex studies, motor unit number estimation (MUNE) procedures, and normalization factors for voluntary electromyographic (EMG) activity. These methodologies assume, with little evidence, that M-wave variability is minimal. This study therefore examined the intraclass reliability of these measures for the biceps brachii. METHODS: Twenty-two healthy adults (4 males and 18 females) participated in 5 separate days of electrical stimulation of the musculocutaneous nerve supplying the biceps brachii muscle. A total of 10 stimulations were recorded on each of the 5 test sessions: a total of fifty trials were used for analysis. A two-factor repeated measures analysis of variance (ANOVA) evaluated the stability of the group means across test sessions. The consistency of scores within individuals was determined by calculating the intraclass correlation coefficient (ICC). The variance ratio (VR) was then used to assess the reproducibility of the shape of the maximum M-wave within individual subjects. RESULTS: The P-P amplitude means ranged from 12.62 ± 4.33 mV to 13.45 ± 4.07 mV across test sessions. The group means were highly stable. ICC analysis also revealed that the scores were very consistent (ICC = 0.98). The group means for the area of the negative phase of the maximum M-wave were also stable (117 to 126 mV·ms). The ICC analysis also indicated a high degree of consistency (ICC = 0.96). The VR for the sample was 0.244 ± 0.169, which suggests that the biceps brachii maximum M-wave shape was in general very reproducible for each subject. CONCLUSION: The results support the use of P-P amplitude of the maximum M-wave as a methodological control in H-reflex studies, and as a normalization factor for voluntary EMG. The area of the negative phase of the maximum M-wave is both stable and consistent, and the shape of the entire waveform is highly reproducible and may be used for MUNE procedures

    Hydrological and associated biogeochemical consequences of rapid global warming during the Paleocene-Eocene Thermal Maximum

    The Paleocene-Eocene Thermal Maximum (PETM) hyperthermal, ~ 56 million years ago (Ma), is the most dramatic example of abrupt Cenozoic global warming. During the PETM surface temperatures increased between 5 and 9 °C and the onset likely took < 20 kyr. The PETM provides a case study of the impacts of rapid global warming on the Earth system, including both hydrological and associated biogeochemical feedbacks, and proxy data from the PETM can provide constraints on changes in warm climate hydrology simulated by general circulation models (GCMs). In this paper, we provide a critical review of biological and geochemical signatures interpreted as direct or indirect indicators of hydrological change at the PETM, explore the importance of adopting multi-proxy approaches, and present a preliminary model-data comparison. Hydrological records complement those of temperature and indicate that the climatic response at the PETM was complex, with significant regional and temporal variability. This is further illustrated by the biogeochemical consequences of inferred changes in hydrology and, in fact, changes in precipitation and the biogeochemical consequences are often conflated in geochemical signatures. There is also strong evidence in many regions for changes in the episodic and/or intra-annual distribution of precipitation that has not widely been considered when comparing proxy data to GCM output. Crucially, GCM simulations indicate that the response of the hydrological cycle to the PETM was heterogeneous – some regions are associated with increased precipitation – evaporation (P – E), whilst others are characterised by a decrease. Interestingly, the majority of proxy data come from the regions where GCMs predict an increase in PETM precipitation. We propose that comparison of hydrological proxies to GCM output can be an important test of model skill, but this will be enhanced by further data from regions of model-simulated aridity and simulation of extreme precipitation events

With its professional roots in late 19th-century industrial Lancashire, stadiums built for the professionalization of football first appear in frequency in the North of England. While many historians of sport focus on consumerism and ‘topophilia’ (attachment to place) regarding these local football grounds, archaeological research that has been conducted on the spectator experience suggests status differentiation within them. Our excavations at Peel Park confirm this impression while also showing a significant afterlife to this stadium, particularly through children’s play

    Multi-modality detection of SARS-CoV-2 in faecal donor samples for transplantation and in asymptomatic emergency surgical admissions

    Background: Faecal transplantation is an evidence-based treatment for Clostridioides difficile. Patients infected with SARS-CoV-2 have been shown to shed the virus in stool for up to 33 days, well beyond the average clearance time for upper respiratory tract shedding. We carried out an analytical and clinical validation of reverse-transcriptase quantitative (RT-qPCR) as well as LAMP, LamPORE and droplet digital PCR in the detection of SARS-CoV-2 RNA in stool from donated samples for faecal microbiota transplantation (FMT), spiked samples and asymptomatic inpatients in an acute surgical unit.  Methods: Killed SARS-CoV-2 viral lysate and extracted RNA was spiked into donor stool & FMT and a linear dilution series from 10-1 to 10-5 and tested via RT-qPCR, LAMP, LamPORE and ddPCR against SARS-CoV-2. Patients admitted to the critical care unit with symptomatic SARS-CoV-2 and sequential asymptomatic patients from acute presentation to an acute surgical unit were also tested. Results: In a linear dilution series, detection of the lowest dilution series was found to be 8 copies per microlitre of sample. Spiked lysate samples down to 10-2 dilution were detected in FMT samples using RTQPCR, LamPORE and ddPCR and down to 10-1 with LAMP. In symptomatic patients 5/12 had detectable SARS-CoV-2 in stool via RT-qPCR and 6/12 via LamPORE, and in 1/97 asymptomatic patients via RT-qPCR. Conclusion: RT-qPCR can be detected in FMT donor samples using RT-qPCR, LamPORE and ddPCR to low levels using validated pathways. As previously demonstrated, nearly half of symptomatic and less than one percent of asymptomatic patients had detectable SARS-CoV-2 in stool
